In your Creator Dashboard, open the emotes page and upload the three sizes from the twitch/ folder (28, 56 and 112 pixels), or the animated version if your channel has access to it.

Server Settings, Emoji, Upload Emoji: take the 128 PNG from discord/, or the GIF from anime/ for an animated emoji (depending on your server's level).

The 512 WebP in grand/ is for graphics, thumbnails and break screens.
